Recombinant Human Collagen Type III in Mimetic Coating of Cardiovascular Stents

Collagen is a core component of the extracellular matrix (ECM) and has been widely used in tissue engineering. However, the inherent thrombogenic properties of collagen hinder its use in blood contact devices. Yang ^[1] et al. reported a novel recombinant human type III collagen (hCOLIII) in which the adhesive sequence contains GER and GEK, without hydroxyproline. It does not have platelet-binding sites while retaining its affinity for endothelial cells. They found that hCOLIII-based ECM-mimetic coating can act as a tailored blood-contacting material for cardiovascular stents.

At first the substrate (Si or PLA) and the PLA stents with a substrate-independent amine-rich polydopamine coating are pre-coated, and then soaked the pre-treated scaffold in hyaluronic acid (HA) solution and hCOLIII solution for multiple cycles to form a multilayer HA/hCOLIII coating. Finally, its application in cardiovascular stent implantation was verified. Platelet adhesion/activation and ex vitro antithrombotic tests confirmed the excellent blood compatibility of the ECM-mimetic coating. In vivo stent implantation data demonstrated that ECM coatings have the ability to promote in situ endothelialization and suppress the excessive neointima hyperplasia, providing a new avenue for the development of functional coatings for cardiovascular scaffolds with collagen-based materials.
