This study focuses on the neurotoxic and genotoxic effects of sublethal doses of acetamiprid on Drosophila melanogaster and Drosophila suzukii over multiple generations, highlighting DNA damage and impaired locomotion as key concerns.
Methodology: Multigenerational cultures of D. melanogaster and D. suzukii were raised on media containing sublethal concentrations of acetamiprid and nicotine. Neurotoxic and genotoxic effects were assessed through climbing tests to measure locomotion and genetic analyses to detect DNA damage in neuroblasts. The persistence of these effects across generations was also examined.
Results: Both species exhibited significant DNA damage in neuroblasts at sublethal acetamiprid concentrations. D. suzukii was more sensitive to the insecticide, showing greater neurotoxic effects compared to D. melanogaster. Behavioral tests indicated reduced mobility in both species, which persisted across generations. This chronic exposure also led to resistance to neonicotinoids over time, potentially compromising long-term pest control efficacy.
Conclusion: Even at sublethal concentrations, acetamiprid caused DNA damage and impaired locomotion in Drosophila species, with D. suzukii exhibiting heightened sensitivity. These findings underscore the need for re-evaluating the long-term ecological impact of neonicotinoids, particularly concerning their genotoxicity and the risk of pest resistance under chronic low-dose exposure.
