MO-TMS Derivatization of Tetrahydrocorticoids for GC-MS Analysis

The derivatization of tetrahydrocorticoids into methoxime-trimethylsilyl (MO-TMS) derivatives represents a reliable method for their quantitative analysis via gas chromatography-mass spectrometry (GC-MS).
Methodology: Tetrahydrocorticoids (approximately 1 mg) were subjected to a sequential derivatization procedure. Initially, 100 μL of methoxyamine hydrochloride in pyridine (5%, w/v) was added to the residue in a 0.3-mL microproduct V-vial. The reaction mixture was sealed under a nitrogen atmosphere and heated to 100°C for 30 minutes, enabling the formation of methoxime derivatives. Following solvent removal under a nitrogen stream, 50 μL of N,O-bis(trimethylsilyl)acetamide (BSA) was introduced to the residue. The reaction mixture was resealed under nitrogen and heated at 100°C for 2 hours to facilitate trimethylsilylation. Post-reaction, the mixture was cooled, and the solvent was evaporated under nitrogen. A subsequent addition of 200 μL of n-hexane enabled preparation for silica cartridge purification. The solution was passed through a Sep-Pak silica cartridge, eluted with 7 mL of n-hexane. The eluate was evaporated at room temperature under nitrogen to yield the tetrahydrocorticoids as MO-TMS derivatives, suitable for GC-MS analysis.
