This work reported the anti-enteroviral activity of the natural anticancer compound OSW-1. OSW-1 potently inhibits the replication of all enteroviruses tested, acting at the genome replication stage, and is effective in all tested cell types from three different species. Importantly, oxysterol-binding protein (OSBP) overexpression rescued viral replication, indicating that the antiviral effect of OSW-1 is due to targeting OSBP.
Evaluation of anti-enteroviral activity of OSW-1
· The effectiveness of OSW-1 was assessed against single rounds of infection in HeLa cells and buffalo green monkey (BGM) kidney cells using various viruses from different species within the Enterovirus genus. These included enterovirus 71 (EV71, part of enterovirus A), coxsackievirus A21 (CVA21, belonging to enterovirus C), human rhinovirus 2 (HRV2, classified as rhinovirus A), and human rhinovirus 14 (HRV-14, categorized under rhinovirus B).
· All tested enteroviruses were found to be sensitive to OSW-1, with IC50 values ranging from 2.4 to 9.4 nM. Parallel cell viability assays indicated no cytotoxic effects (CC50 > 100 nM) during the 10-hour infection period. Additionally, coxsackievirus B3 (CVB3, of enterovirus B) with Renilla luciferase expression (RLuc-CVB3) was also effectively inhibited by OSW-1.
· The study further examined OSW-1's impact on the replication of other picornaviruses. While equine rhinitis A virus (ERAV), from the Aphthovirus genus, showed slight inhibition at higher concentrations of OSW-1, those concentrations also compromised cell viability after 24 hours, which was when virus replication was evaluated. In contrast, human parechovirus 1 (HPeV1) from the Parechovirus genus demonstrated no inhibition.
OSW-1 exhibits potent anticancer activity in various cancers. This work summarized the available evidence for the anticancer effects and mechanisms of OSW-1 in vitro and in vivo. OSW-1 exerts its anticancer effects through multiple mechanisms, the core of which include inhibition of cancer cell proliferation, induction of apoptosis, and activation of the Golgi stress response.
· OSW-1 is able to inhibit cancer cell proliferation, and it does so by interfering with the cell cycle, specifically by causing cell cycle arrest in HL-60 cells at the G2/M phase when treated with 0.3 nM or 0.01 µM. In addition, OSW-1 was found to exert antiproliferative activity by binding to alcohol oxygen binding protein (OSBP) and its related proteins (ORPs), and ORP4 is considered to be the main antiproliferative target of OSW-1.
· OSW-1 effectively induces apoptosis in cancer cells, and this process is mainly achieved through the mitochondria-mediated intrinsic pathway and the death receptor-mediated extrinsic pathway. OSW-1 can promote apoptosis in different types of cancer (such as breast cancer, colon cancer and leukemia), often by activating caspase-3 and other apoptosis-related proteins; in addition, OSW-1 may also increase apoptosis through a Golgi stress-induced mechanism.
· OSW-1 can activate the Golgi stress response, which involves multiple signaling pathways. OSW-1 can cause the mislocalization of OSBP in the cell Golgi network, thereby activating the TFE3 and CREB3-ARF4 pathways, which are closely related to the apoptosis process.
