4-Aza-dl-leucine dihydrochloride (AZL) is a leucine analog that can be used as a resistance screening compound in metabolic engineering studies to obtain compounds with high 3-methyl-1-butanol Yields feedback inhibition of AHAS or IPMS mutants. AZL has been previously used against Bacillus subtilis, Escherichia coli, and Salmonella typhimurium to identify drug-resistant strains.
Mutagenicity Experiment
· Yeast cells were adjusted to pH 6.0 with phosphate buffer solution. The chemical mutagen 1-methyl-3-nitro-1-nitroso-guanidine (NTG) was dissolved in 1 mL of acetone and added to each sample (5 mL).
· Adjust the final concentration of NTG to 500 μg/mL, and then incubate in a constant temperature shaker at 30°C for 15 min. The mutant cells were then collected by centrifugation and washed three times with distilled water.
· 200 μL of resuspended mutant cells were inoculated on yeast nitrogen base (YNB) solid medium containing 20 mg/mL 4-Aza-dl-leucine dihydrochloride (AZL), and then cultured at a constant temperature of 30°C. Incubate in the box for about three days until colonies appear.
· Finally, fermentation experiments were conducted using fermented glucose and duckweed hydrolyzate, and the auxotrophic strain with the highest C4~C5 alcohol concentration was identified.
