· The pharmacological mechanisms: Sal B works by inhibiting the expressions of Tumor necrosis factor (TNF)-α, Cyclooxygenase (COX2), and NADPH oxidase (NOS) through the inhibition of TNF-α/Nuclear factor κB (NF-κB) and Toll-like receptor (TLR4)/NF-κ-B pathways. It also enhances the expression of antioxidant enzymes like Heme oxygenase-1 (HO-1), NQOD, and Superoxide dismutase (SOD), while inhibiting the expression of oxidases such as NOX-2, NOX-4, and Poly (ADP-ribozyme) polymerase 1 (PARP-1) through the Nuclear factor E2-related factor 2/Kelch-like ECH-related protein 1 pathway. This ultimately leads to the inhibition of Reactive oxygen species (ROS) generation and improvement in inflammation, cell apoptosis, autophagy, fibrosis, microcirculation disorders, and stem cell proliferation and differentiation.
· Sal B exerts its mechanisms in various chronic inflammatory diseases by increasing the expression of antioxidant enzymes like Superoxide dismutase (SOD), CAT, and GSH, and decreasing the expression of oxidative enzymes such as Cyclooxygenase (COX-2), NADPH oxidase (NOX)-4, and iNOS. This inhibition of Reactive oxygen species (ROS) and malondialdehyde (MDA) generation contributes to the improvement of chronic inflammation in different disease models. The findings suggest a close relationship between the anti-inflammatory and antioxidant effects of Sal B in treating chronic inflammatory diseases.
