Amphiphilic diblock copolymers consisting of methoxypolyethylene glycol (MePEG) and '-caprolactone ('-CL) were prepared by MePEG-initiated '-CL polymerization. MePEG/'-CL block copolymeric micelles containing indomethacin (IMC) were prepared by dialysis and evaluated as novel drug carriers. The size of the formed micelles was less than 200 nm, and the micelle size distribution showed a narrow and monodisperse unimodal pattern. In addition, the micelles formed by the dialysis method exhibited a spherical structure. For those prepared using copolymers with a molecular weight of about 12 000 and a polymer/IMC weight ratio of 1/1, the content of indomethacin in the nanospheres was about 42.2%. The release rate of indomethacin from the nanospheres was slow, so that the release was sustained for more than 15 days. The release rate decreased with the increase of the copolymer molecular weight and the amount of encapsulated drug. These results indicate that the drug-loaded nanospheres can be used as novel drug carriers in hydrophobic drug injection delivery systems. ,
MePEG/'-CL diblock copolymers were prepared by polymerization of '-caprolactone to remove unbound IMC and aggregated IMCMePEG, the obtained micellar solution was sonicated, centrifuged and tested with various copolymer compositions in Table 1, and then lyophilized. Subsequently, the nanospheres were edited. The "MEP series" was obtained by freeze-drying micelles with a fixed molecular weight of 5000 in the MePEG chain and the PCL part was broken from M/I535 to 150 by adding ethanol and THF. MePEG/'-CL (1:1 v/v), the amount of IMC entrapped was the deblocked copolymer containing IMC. MePEG/'-CL block copolymer (100 mg) was dissolved in 10 mL of the '-caprolactone part of the nanospheres in 10 mL of selected organic solvents, which were determined according to the weight of the nanospheres and the amount of drug loaded such as dimethylformamide (DMF), tetrahydrofuran (THF), dimethylacetamide (DMA) and dichloromethane from the following equation.
