A study on the effects of virgin coconut oil (VCO) on preventing blood pressure increases and improving endothelial function was conducted on rats fed with repeatedly heated palm oil. This study investigated the effects of VCO on blood pressure, plasma nitric oxide levels, and vascular reactivity in male rats repeatedly fed with heated palm oil. In his study, repeated feeding of heated palm oil resulted in an increase in blood pressure. When overheated, the free radicals produced cause oxidative stress in the blood vessels, affecting nitric oxide levels in endothelial cells. In male rats, supplementation with repeatedly heated palm oil VCO was found to prevent blood pressure increases and reduce nitric oxide inactivation. In addition, VCO did not affect vasodilation, but reduced endothelial vasoconstriction.
Virgin coconut oil (VCO) with a high polyphenol content is able to maintain normal levels of cholesterol and other lipid parameters in tissues and serum, and also increases the concentration of high-density lipoprotein cholesterol in rats. The antioxidant activity of the polyphenol fraction (PF) of VCO against copper-induced low-density lipoprotein (LDL) oxidation confirms the great potential of this oil in protecting LDL from oxidative stress induced by physiological oxidants.
The animals were housed individually in polypropylene cages, the room temperature was maintained at 25°F 1°C, and there was a 12-h light and 12-h dark cycle. The experimental groups were as follows: Group I (control) - peanut oil (GNO), Group II - coconut oil CO (8%), Group III - VCO (8%). The oils were fed together with semi-synthetic feed for 45 days. 2 ml of serum was thoroughly mixed with 0.2 ml of heparin-manganese solution (final concentrations of 1.4 mg/ml and 0.092 M, respectively). The samples were incubated at room temperature for at least 10 min and centrifuged at 1500 g for 30 min. The clear supernatant was removed manually for the assay. HDL-C was measured as described by Abell. LDL and VLDL cholesterol levels were calculated using the standard Friedewald equation. PF was extracted from the test oils according to the method described by Vazquez Roncero et al. 10 g of the oil was dissolved in 50 ml of hexane and extracted three times with 20 ml of 60% methanol in sequence. The vacuum-dried final residue obtained from the combined extracts was dissolved in a known volume of methanol. The total polyphenol content of this solution was estimated using the Folin-Ciocalteau reagent.
