2-Methacryloyloxyethyl Phosphorylcholine for the Synthesis of Protein Delivery Nanocapsules

Protein therapeutics with stealth capabilities can be successfully engineered by coating individual protein molecules with thin layers of zwitterionic polymers to resist nonspecific protein uptake. In this strategy, phosphocholine (PC) polymer nanocapsules were synthesized to extend the circulation time of the therapeutic protein and minimize its immunogenicity. In this work, 2-methacryloyloxyethyl phosphorylcholine (MPC), as a monomer, is an important synthetic raw material for PC polymer nanocapsules.
Synthesis of nanocapsules with MPC
· Proteins were first pre-conjugated with polymerizable acryloyl groups using N-hydroxysuccinimide ester, and encapsulated via in situ polymerization at room temperature using MPC as the monomer, BIS as the crosslinker, and ammonium persulfate and tetramethylethylenediamine as the initiator.
· In the synthesis procedure, solutions of MPC and BIS were created as 10% (m/v) stock solutions in DI-water and anhydrous DMSO, respectively. These solutions were then mixed with the proteins at a specific molar ratio. The protein concentration was adjusted to 1 mg/mL with a phosphate buffer (50 mM, pH 7.0). Polymerization was initiated by adding APS and TEMED, followed by keeping at 4 °C for 2 hours.
