Quantitative Determination of Fipronil and Its Intermediates in Soil

Fipronil is an insecticide and the active ingredient in many formulations. When applied to crops, fipronil may undergo chemical or biodegradation to produce intermediates. In order to detect these persistent fipronil intermediates, this work used extraction combined with ultrasound and GC-ECD to simultaneously quantify fipronil and its intermediates including fipronil desulfinyl, fipronil sulfone and fipronil sulfide.
Quantitative method
· The analytes were extracted using an ultrasonic bath and the solvent ratio, number of cycles, and cycle time were optimized through a Box-Behnken design with a triplicate central point. Optimal extraction conditions were determined using response surface analysis.
· A clean-up step was performed using solid-phase extraction (SPE) cartridges containing silica and aluminum oxide. Gas chromatography with electron capture detection (GC-ECD) was utilized to separate fipronil and its intermediates (including fipronil sulfide) with a resolution suitable for a 20-minute runtime. The best quantification results were obtained using a 1:1 (v/v) acetone/hexane ratio and 2 ultrasound cycles of 15 minutes each.
· Recovery values ranged from 81 to 108%, with a relative standard deviation (RSD) below 6% and no impact from the matrix used. Analytical curves showed regression coefficients above 0.9908 for concentrations ranging from 0.005 to 0.6 μg g-1. Limits of detection (LOD) ranged from 0.002 to 0.006 μg g-1 and limits of quantification (LOQ) ranged from 0.006 to 0.020 μg g-1 for all analytes.
