Delmopinol has demonstrated significant potential in dental care applications, particularly when combined with fluoride. This study explored the effects of delmopinol and fluoride, individually and synergistically, on acid adaptation and acid production in plaque biofilm bacteria under in vitro conditions.
Biofilm bacteria were exposed to delmopinol (0.16 mM), fluoride (1 mF NaF), or their combination under static conditions at pH 5.5 to assess acid adaptation. The following day, the bacteria were challenged with an acidic environment (pH 2.5) to evaluate acid tolerance, using confocal scanning laser microscopy and LIVE/DEAD BacLight Viability staining to quantify viable cells. Acid production was measured via pH drop after glucose pulsing with various concentrations of delmopinol and fluoride.
The findings revealed that fluoride alone significantly reduced acid tolerance and acid production. Notably, a combination of delmopinol and fluoride had a more pronounced inhibitory effect on acid adaptation and acid production, even at concentrations where the individual compounds were ineffective. Delmopinol alone did not impact acid adaptation or acid production.
