Ceftazidime (CAZ) is extensively prescribed for treating Acinetobacter baumannii-induced infections, particularly nosocomial pneumonia in immunocompromised patients. A series of two-fold dilutions (16 mg/mL to 15 μg/mL) were tested against A. baumannii KSK1 in triplicates, and the lowest concentration preventing visible bacterial growth after 24 hours at 37 °C was designated as the MIC. Sub-MIC concentrations demonstrated multiple impacts: (1) reduced adhesion to lung epithelial cells, slowing bacterial growth temporarily; (2) altered bacterial cell morphology, leading to aggregation resembling biofilm structures; and (3) promoted in vitro biofilm formation, a key factor in bacterial persistence and resistance.
The findings highlight CAZ's dual role: while effective against A. baumannii at therapeutic concentrations, sub-MIC levels in environmental settings, such as wastewater, may inadvertently select for biofilm formation and antibiotic resistance.
