Semduramicin (Aviax) is an ionophore coccidiostat licensed as a fattening feed additive for chickens. In order to determine whether feed samples comply with legal requirements and to check whether the labeling is correct, an efficient method based on liquid chromatography, UV spectrophotometric detection and post-column derivatization (LC-UV-PCD) was proposed.
Preparation of test samples containing semduramicin
· Compound feed samples: Specifically, 25 ± 0.1 g of the compound feed samples were spiked with varying amounts of Aviax 5%. These amounts were chosen to align with the authorized concentration of semduramicin in the feed additive, resulting in concentrations of 12.8 mg kg-1, 25.7 mg kg-1, and 51.3 mg kg-1. Additionally, the samples were fortified with a mixture of other coccidiostats to ensure proper penetration into the feed matrix before further analysis.
· Premixtures: In another set of experiments, premixture samples that did not contain semduramicin were mixed with Aviax 5%, leading to a dilution of the target compound. To validate the process, these premixture samples were spiked with appropriate amounts of Aviax 5% to achieve a target concentration of semduramicin of 3.07 g kg-1 in the premixture.
· Feed additive: As for the feed additive Aviax 5%, it was used as is without any additional treatment. The specific target concentration of semduramicin in the feed additive was 51.3 g kg-1 according to the relevant regulations.
