Biosynthetic Pathway for Pheromones Such as (Z)-9-Tetradecadienyl Acetate

(Z, E)-9,12-tetradecadienyl acetate (ZETA,Z9,E12-14:OAc) is the major sex pheromone component of many stored food moth species. This pheromone is used worldwide for disruption, detection, monitoring, and large-scale trapping of fresh and processed food storage facilities. In this study, the bioproduction of the ZETA pheromone by the engineered yeast Saccharomyces cerevisiae was demonstrated, which also involved the production of (Z)-9-tetradecene acetate (Z9-14:OAc).
The biosynthesis of Z9,E12-14:OAc in E. cautella has been shown as figure. To create a biosynthetic pathway for ZETA production, this study utilized Dmel_D9 from D. melanogaster. The pathway was assembled using SexipgFARII38 and yeast ATF139. SexipgFARII effectively reduces Z9,E12-14:Acyl, while ATF1 is highly efficient at acetylating (Z, E)-9,12-tetradecadien-1-ol (Z9,E12-14:OH).
When the complete biosynthetic pathway was expressed in yeast and myristic acid methyl ester (14:Me) was added to the yeast medium, the final pheromone components, Z9,E12-14:OAc and (Z)-9-tetradecenyl acetate (Z9-14:OAc), were detected in the medium after four days of cultivation.
