Mechanism Of Cis-Nerolidol-Induced Death In Cancer Cells

Nerolidol is a naturally occurring sesquiterpene alcohol with multiple properties, including antioxidant, antimicrobial, and antiparasitic activities. In this study, the researchers investigated the antitumor mechanism of cis-nerolidol in bladder cancer cell lines. The results of experiments in two bladder cancer cell lines showed that nerolidol inhibited cell proliferation and induced two different cell death pathways. The researchers confirmed that cis-nerolidol induces DNA damage and ER stress. Cis-nerolidol shows promising antitumor activity through an unorthodox mechanism of action and can help target drug-resistant malignancies such as bladder cancer.
Cells were seeded into 24-well plates or 12-well plates and grown overnight. Three treatments were performed with different concentrations of cis-nerolidol (25, 50, 60, 75, and 10 0 mg/L) specified for each experiment. Incubation time was 0.5, 2, 4, 24, 48, or 7 2 h of treatment as indicated in each experiment. For experiments involving inhibitors, cells were pretreated with inhibitors for 1 h before administration of cis-Nerolidinone. Cell counts were performed using a microscope using a Neubauer chamber. After obtaining a single-cell suspension, cell viability was assessed by staining the cells with trypan blue stain. Cells were counted as described previously. Cells stained blue were considered nonviable. Data are expressed as the proportion of trypan blue-stained cells to the total cell count (100%) for each measurement.
