Ammonium trichloro[1,2-ethanediolato-O,O']-tellurate (AS101) is an important bioactive synthetic tellurium compound that can serve as an effective immunomodulator and exhibits a variety of potential Therapeutic applications and anti-tumor effects. A novel microwave-assisted synthesis (MAOS) strategy has been developed for the synthesis of AS101, which has the advantage of shorter reaction times.
Synthesis methods of AS101
· Method A: AS101 was synthesized by refluxing Te(IV) and ethylene glycol in acetonitrile. TeCl4 (0.675g) and ethylene glycol (0.387g) were dissolved in acetonitrile (5mL). The mixture was microwaved at 50W for 30 minutes. After cooling the reaction mixture, a white crystalline solid was obtained, washed with acetonitrile, filtered and dried under vacuum.
· Method B: AS101 was synthesized by refluxing Te(IV) and ammonium chloride in ethylene glycol. TeCl4 (1.35g) and NH4Cl (0.387g) were dissolved in ethylene glycol (5 mL). The mixture was microwaved at 80W for 10 minutes. After cooling the reaction mixture, a white crystalline solid was obtained, washed with acetonitrile, filtered and dried under vacuum.
The small tellurium IV compound, trichloro(dioxyethylene-o,o')ammonium tellurate (AS101), is a potent in vitro and in vivo immunomodulator with multiple potential therapeutic applications. AS-101 has a variety of biological activities, such as anti-inflammatory activity, antiviral activity, chemosensitization of cancer cells, and prevention of chemotherapy induced toxicity. The mechanism of AS-101's anti-autoimmune activity has been demonstrated in a variety of autoimmune diseases, including inflammatory bowel disease (IBD), multiple sclerosis (MS), and psoriasis.
Effects of AS-101 on autoimmune diseases
· The non-toxic, small tellurium IV compound, AS101, exerts anti-inflammatory/autoimmune activity in various experimental autoimmune diseases and in patients.
· The anti-autoimmune properties of AS-101 are mediated by several multifunctional activities that include A) its ability to inhibit Th17/ IL-17 function; B) its specific unique redox-modulating activities, enabling the inhibition of specific leukocyte integrins (α4β1 and α4β7); C) its ability to restore the Treg population and D) its ability to induce IL-2 production and thereby affect Th17/Treg balance.
· The anti-autoimmune activities of AS-101 suggest that tellurium-based compounds may be promising candidates for the management of autoimmune diseases.
