Effects of RH5849 and Mifepristone on Female Drosophila Lifespan

Ecdysone is a Drosophila steroid hormone reported to regulate mating responses. 1,2-Dibenzoyl-1-(T-Butyl)Hydrazine (RH5849) is a synthetic ecdysone agonist and a potent mimetic of ecdysone. Studies have explored the lifespan effects of RH5849 on virgin females fed 1000 µg/ml in the presence and absence of 200 µg/ml mifepristone. The results are as follows:
· Administration of 200 µg/ml mifepristone alone led to an expected increase in life span (+23.5% and +26.1%, respectively).
· Conversely, treatment with 1000 µg/ml RH5849 alone resulted in a significant decrease in life span (-79.4% and -78.5%, respectively).
· However, when co-treated with 200 µg/ml mifepristone, a substantial rescue in life span was observed (+136% and +157%, respectively).
· To eliminate the possibility that the increased life span from mifepristone was due to reduced food intake, the EXQ assay was used to measure food consumption. Administration of 1000 µg/ml RH5849 led to a decrease of approximately 75% in food intake, while mifepristone did not impact food intake, suggesting that the rescue in life span by mifepristone was not a result of decreased food or drug intake.
