Procedure for The Extraction and Purification of Verbascoside
Sample Preparation:
- If using fresh plant material, freeze it in liquid nitrogen and grind to a fine powder using a mortar and pestle.
- If using dried plant material, grind it to a fine powder using a mortar and pestle.
Extraction:
- Weigh 10 grams of the prepared plant material and transfer it to a conical flask.
- Add 100 mL of methanol to the flask and seal it tightly.
- Place the flask in an ultrasonic bath and sonicate for 30 minutes at room temperature.
- Filter the extract through a filter paper or a syringe filter to obtain a clear solution.
Purification:
- Transfer the filtered extract to a separating funnel.
- Add an equal volume of ethyl acetate to the separating funnel.
- Shake vigorously for several minutes to ensure efficient partitioning.
- Allow the layers to separate, and carefully drain the lower ethyl acetate layer into a separate container.
- Repeat the extraction with ethyl acetate twice more to ensure maximum recovery.
Quantification of Verbascoside
Verbascoside is usually quantified using high-performance liquid chromatography (HPLC) analysis: Prepare a standard solution of verbascoside by dissolving a known amount of verbascoside in methanol to obtain a desired concentration range. Inject the standard solution and the purified sample extract into an HPLC system equipped with a suitable column and detector (e.g., C18 column and UV detector). Set the HPLC conditions, including the mobile phase composition, flow rate, and detection wavelength, optimized for verbascoside analysis. Inject a defined volume of the sample extract into the HPLC system and record the peak area or height. Calculate the verbascoside concentration in the sample using the calibration curve generated from the standard solutions.
