The aminoglycoside antibiotic neomycin is used to treat external or internal bacterial infections and is mainly used in veterinary medicine in the form of its sulfate salt. This work investigated the pharmacokinetic characteristics and absolute availability of neomycin sulfate in swine after a single intravenous and oral dose of 15 mg/kg body weight.
Evaluation methods and results
The blood samples were analyzed using ultra-high-performance liquid chromatography coupled with tandem mass spectrometry (UPLC-MS/MS), and the pharmacokinetic parameters were evaluated through a non-compartmental approach.
In swine, following oral administration, the elimination half-life, mean residence time from the initial time to the last collection point, mean maximum concentration, mean time to reach maximum concentration, and the area under the concentration-time curve from the initial time to the last collection point were recorded as 12.43 ± 7.63 hours, 10.25 ± 4.32 hours, 0.11 ± 0.07 μg/ml, 1.92 ± 0.97 hours, and 1.23 ± 0.78 μg·h/ml, respectively. In contrast, after intravenous administration, these values were 5.87 ± 1.12 hours, 6.07 ± 0.49 hours, 15.80 ± 1.32 μg/ml, 0.30 ± 0.38 hours, and 76.14 ± 3.52 μg·h/ml. The absolute bioavailability of neomycin sulfate B was determined as 4.84% ± 0.03.
In summary, the pharmacokinetic profile indicated a long elimination half-life and a slow clearance rate. The low absolute bioavailability of neomycin sulfate following oral administration suggests that future research could enhance its absorption for treating systemic infections by modifying the dosage form or incorporating adjuvants.
