TCO

TCO are a class of click chemistry reagents containing a reactive TCO group. As dienophiles, TCO can react with tetrazines by inverse electron-demand Diels-Alder (IEDDA) click reaction, which is the fastest bioorthogonal reaction found so far, and the reaction rate can reach 10000 under physiological conditions without catalysts. Due to the high reaction rate and good biocompatibility, IEDDA click reaction is widely used in labeling and bioconjugation. The structure of TCO group is shown in scheme 1.
